From Santa Cruz you can take a bus to Sucre but it is a long and arduous bus trip of approximately 12 hours through the mountains with few stops for food and rest. You would probably prefer to fly. Flights from Santa Cruz to Sucre take under one hour and cost less than $100 (usually about $75 or $80) one way. From Sucre you can take a taxi or bus to Potosí (about 4 hours). Potosí does not yet have a commercial airport so you have to go through Sucre to get to the city of Potosí.
If you are planning a trip to Uyuni, there is a tiny airport in Uyuni to which very small planes are flying, but they are only a few times a week and they leave from Cochabamba, if I'm not mistaken. The airport in Uyuni is not a commercial airport, it is only for very tiny planes.
